TASP1 (taspase1) - the protease that cuts KMT2A, and the timer the fusion escapes
also called taspase1 · threonine aspartase 1 · TASP1
Taspase1 is the enzyme that cuts normal KMT2A in half. The leukemic fusion protein is missing the places where the cut happens, so it is never cut - and that turns out to be one reason it lasts so long in the cell.
The same thing, for a scientist
Taspase1 is a threonine aspartase that cleaves after aspartate using an N-terminal threonine nucleophile, and is itself intramolecularly processed into an active 28 kDa alpha / 22 kDa beta heterodimer [E1]. Its identified mammalian nuclear substrates are MLL1, MLL2 and TFIIA [E2]. It cleaves KMT2A at D2666 and D2718, generating N320 and C180. What that cleavage does is contested: Takeda et al. read Taspase1-null mice as showing that cleavage activates MLL, with reduced precursor HMT activity and lost H3K4me3 at Cyclin E1/E2 [E3]; Zhao et al. read taspase1 knockouts in human cells as showing that cleavage primes MLL1 for proteasomal degradation, with the uncleaved protein longer-lived and markedly more chromatin-bound [E2]. Both sides matter to this graph because **no der(11) KMT2A fusion contains either cleavage site**. As a drug target taspase1 itself looks poor: a 30-million-compound virtual screen produced 26 candidates and none worked, and 70% knockdown does not change substrate cleavage [E2]. The tractable node is the kinase that licenses the cleavage, CKII.
Why this protein has an entry in a fusion-oncoprotein graph
Because it is a regulator that acts exclusively on the allele the fusion is not. Taspase1 cannot touch any der(11) KMT2A fusion - the sites are ~1,300 residues past the breakpoint - yet manipulating it changes what the fusion does, by changing how much wild-type KMT2A is on chromatin competing with it 2. It is the clearest worked example in this disease of a regulatory circuit that is structurally unable to reach the oncoprotein and is nonetheless a lever on it.
It is also the reason the taspase1 sites moved from 'no demonstrated consequence' to a demonstrated one in ana-kmt2a-lost-module-ledger during the cycle-31 correction pass.

| relation to fusion | cannot act on any der(11) KMT2A fusion - both sites are ~1,300 residues C-terminal to the breakpoint cluster region; acts only on the retained wild-type allele and on the reciprocal der(4) AFF1::KMT2A product |
| druggability | poor as a direct target: a 30-million-compound virtual screen yielded no working inhibitor, and 70% knockdown does not change substrate cleavage (PMID 30573454). The tractable node is CKII, whose inhibition blocks the licensing phosphorylation on the substrate. |
| contested | whether cleavage activates MLL1 (PMID 16951254) or primes it for degradation (PMID 30573454); a third report of non-cleavable knock-in mice with no phenotype is cited by the latter |

| localization | NUCLEUS, predominantly, with accumulation at the NUCLEOLUS -- shown for endogenous AND ectopically expressed Taspase1 [E_k54_loc1]. Import is active and required for function: an evolutionarily conserved bipartite NLS at residues 197-220 (KRNKRKLELAERVDTDFMQLKKRR) binds importin-alpha, and an NLS-mutated import-deficient Taspase1 is BIOLOGICALLY INACTIVE. A transient cytoplasmic pool exists and is functional: Taspase1 appears to exploit nucleophosmin's nuclear export activity to reach its cytoplasmic substrates. |
| subnuclear localization | NUCLEOLUS, and it is a consequence of activation rather than a resting address [E_k54_loc1]. Nucleolar localization REQUIRES autoproteolytic processing, which triggers interaction with the nucleolar shuttle protein nucleophosmin (NPM1); (auto)catalytically inactive Taspase1 mutants neither accumulate at the nucleolus nor bind nucleophosmin. So the subnuclear address reports the enzyme's activation state, which makes it a readout and not merely a coordinate. |
| localization shown for fusion | NO. Both records are for wild-type Taspase1. Nobody has asked where Taspase1 is in a KMT2A-rearranged cell, and the fusion is not a Taspase1 substrate at the relevant sites anyway -- the cleavage sites D2666 and D2718 lie beyond every leukaemic breakpoint, which is this entry's existing point about the fusion escaping the timer. |
| epigenetic readout | INDIRECT AND ONE PROTEIN AWAY, recorded as MLL's mark rather than Taspase1's. Taspase1 **alters the H3K4 epigenetic signature** upon estrogen stimulation BY CLEAVING MLL, and both estrogen-driven transcription and MLL-derived epigenetic labelling fall on Taspase1 siRNA knockdown [E_k54_top2b]. Taspase1 itself writes, erases and reads no histone mark: it is a threonine aspartase. This is the same filing decision made for gene-top2a and gene-top2b, where the H3K4 change also belongs to MLL and reaches the topoisomerase only through Taspase1. |
| npm1c connection untested here | Coexpression of PATHOLOGICAL NUCLEOPHOSMIN VARIANTS increases the amount of CYTOPLASMIC Taspase1 [E_k54_loc1]. This graph holds NPM1c as a menin-inhibitor-sensitive, CRM1-addressed, HOXA-driving lesion with no KMT2A rearrangement (see mechanism-npm1c-crm1-hoxa-recruitment and the export-axis beat). If NPM1c relocalises the protease that cleaves wild-type MLL1, then an NPM1-mutant blast may process MLL1 differently from a wild-type one -- and nobody has looked. The paper's variants are pathological NPM1 mutants of the class that includes the AML NPM1c lesion, but the paper does not test NPM1c specifically, so this is a lead and not a finding. |
| nls coordinates verified | VERIFIED 2026-09-22 against the sequence, not taken on trust. UniProt Q9H6P5 (primary gene TASP1, 420 aa, matching this entry's length_aa) residues 197-220 are exactly KRNKRKLELAERVDTDFMQLKKRR -- the peptide the paper quotes, in the position it quotes. So the bipartite NLS coordinate is safe to build on, which is not something this project assumes: it stored the PML coiled coil 106 residues short and the KMT2A bromodomain as 1703-1809 when it is 1635-1765, both from not checking a quoted range against the sequence. |

}- Taspase1 is required for normal HOX expression and cell-cycle progression in its own right. Any strategy that works by blocking its activity, rather than by blocking the licensing phosphorylation on one substrate, would hit MLL2 and TFIIA as well - and the mouse knockout is small with proliferation defects.
- If the field's reading of cleavage inverts again, every entry in this graph that describes the fusion as 'skipping maturation' inherits the error. Entries should say what cleavage does mechanistically and cite the dispute, not use the word 'maturation'.
- A Taspase1 inhibitor is not a clean MLL-directed agent. Taspase1 also cleaves TFIIA, and in mice TFIIA is named the PRINCIPAL substrate for craniofacial morphogenesis, acting by limiting p16Ink4a and p19Arf transcription from the Cdkn2a locus (PMID 25664857); Taspase1-null mice have catastrophic craniofacial malformations and acute deletion markedly reduces thymocytes (PMID 16951254). In an infant, a drug whose germline phenotype is craniofacial malformation plus thymic loss is carrying two developmental liabilities that have nothing to do with the fusion.
- Does taspase1 cleavage activate MLL1 or destabilise it? Two careful studies disagree and a third reports no phenotype in non-cleavable mice. This is the single unresolved question that most statements about KMT2A 'maturation' in this graph depend on.
- Which ubiquitin machinery acts downstream of cleavage to degrade MLL1? Zhao et al. explicitly leave it unidentified, and naming it would turn this pathway into a druggable node rather than a descriptive one.
- Has the MLL non-cleavable knock-in allele ever been crossed into a KMT2A-rearranged leukemia model? The tool exists (PMID 25267403) and the pharmacological equivalent worked (PMID 30573454); the genetic experiment appears never to have been reported.
- Does NPM1c relocalise Taspase1 in an AML blast? Pathological nucleophosmin variants increase cytoplasmic Taspase1, and NPM1c is a menin-inhibitor-sensitive lesion in this graph with no KMT2A rearrangement. If the protease that cleaves wild-type MLL1 is redistributed, MLL1 processing may differ between NPM1-mutant and NPM1-wild-type blasts. Nobody has looked.
- Does Taspase1 activity alter KMT2A breakage rates? It enhances TOP2B-mediated double-strand breaks, and TOP2B is the isoform most etoposide-induced KMT2A breaks depend on. Untested in both directions.

Endogenous and ectopically expressed Taspase1 is detected predominantly in the nucleus, accumulating at the nucleolus. An evolutionarily conserved bipartite nuclear import signal at residues 197-220 (KRNKRKLELAERVDTDFMQLKKRR) interacts with importin-alpha, and an NLS-mutated import-deficient Taspase1 is biologically inactive. Nucleolar localisation requires autoproteolytic processing, which triggers interaction with nucleophosmin; (auto)catalytically inactive mutants neither accumulate at the nucleolus nor bind nucleophosmin. Coexpression of pathological nucleophosmin variants increases cytoplasmic Taspase1, and the authors propose Taspase1 exploits nucleophosmin's export activity to reach cytoplasmic substrates.

- Nucleolar accumulation that depends on autoprocessing is exactly the shape an artefact of overexpression takes: a highly expressed, self-cleaving enzyme concentrating in the nucleolus is also what one sees when a protein is aggregating there. The paper reports the endogenous protein too, which is the mitigation, but the mechanistic claims -- the NPM1 interaction and the export route -- rest on coexpression experiments.
- This graph's own registry records that `nucleolar[tiab]` is a contaminated search token and that nucleolar claims here have repeatedly dissolved on inspection -- cycle 49's nucleolar convergence was withdrawn entirely, and MLL-AF4's nucleolar residual is -0.14 pp once bait depth is accounted for. This paper was surfaced by a concurrent beat's registry measurement -- `TOP2A[tiab] AND H3K4[tiab]` and `TOP2B[tiab] AND H3K4[tiab]` both return 0 while `topoisomerase[tiab] AND H3K4me3[tiab]` returns 7 and contains it -- so it was invisible to the obvious query. Evidence that argues against this (1)
- A protease that increases TOP2-mediated double-strand breaks is a mechanistic hazard as well as a mechanism: this graph's break-machinery beat holds that TOP2B is the isoform most etoposide-induced KMT2A breaks depend on. So the same interaction that drives hormone-induced transcription is a candidate route to the very translocation this project studies, and nothing in this paper tests whether Taspase1 activity alters KMT2A breakage rates. That is the opposite of a therapeutic reading and it is untested in both directions.

dominant-negative taspase1 fails in t(4;11) cells because inactive enzyme does not heterodimerise (PMID 22570686)

taspase 1 is the protease whose cleavage makes the N/C difference interpretable
taspase1 is the protease whose cleavage creates the two fragments
lies in the taspase1 N320 chain (1-2718) that the fusion only partly resembles
TASP1 -> TFIIA cleavage -> limited Cdkn2a transcription: non-cleavable TFIIA accumulates at the p16Ink4a and p19Arf promoters and drives them, and p16Ink4a deficiency rescues the TASP1-null craniofacial phenotype (murine; PMID 25664857). An MLL-independent arm of Taspase1 biology.
WEAK: acute Taspase1 deletion markedly reduces thymocytes in mice (PMID 16951254) - one abstract clause with no immunophenotype, so the nearest node is used rather than a measured population
Taspase1 loss down-regulates Cyclins E, A and B, up-regulates p16Ink4a and reduces H3K4me3 at Cyclin E1/E2; MEFs and MLL(nc/nc;2nc/nc) MEFs are both impaired in proliferation (murine; PMID 16951254)
Taspase1 interacts specifically with topoisomerase II alpha and enhances DSB formation for hormone-induced transcription; in the same work Taspase1 alters the H3K4 signature by cleaving MLL
Taspase1 is functionally linked to hormone-induced, topoisomerase-II-beta-mediated transient double-strand breaks; the coupled H3K4 change is deposited by MLL after Taspase1 cleavage
