MBTD1 - the H4K20me1/me2 reader inside NuA4/TIP60, the quiescence brake on adult haematopoietic stem cells, and the 3' partner of ZMYND11::MBTD1 whose methyl-lysine cage the fusion carries and does not use
also called MBTD1 · MBT domain-containing protein 1 · mbt domain containing 1 · SA49P01 · FLJ20055
MBTD1 is a protein that recognises one particular chemical tag on packaged DNA and sits inside a machine that loosens the packaging. In healthy bone marrow it keeps blood stem cells resting, and in a rare leukaemia its back half is welded to another gene to drag that loosening machine to the wrong places.
The same thing, for a scientist
**Verbatim coverage verdict before writing:** `MBTD1 no match under 5 candidate names` from `scripts/is-symbol-covered.py MBTD1` on 2026-09-23 (2331 ids from 2331 node files; HGNC rows 45083, retired/alias index 58536; `covered 0, DECLINED on purpose 0, no match 1, unresolvable 0, total 1`). Word-boundary naming count before this entry: **2 node files**, both written by Q119 last cycle. No row in `data/symbol-collisions.json` (208 rows) for `MBTD1` or for either alias. **Identity, fetched.** Q05BQ5 (`MBTD1_HUMAN`), **628 aa**, `ENSG00000011258`, **HGNC:19866**, cytoband **17q21.33**, GRCh38 chr17:51,177,419-51,261,114 on the minus strand, RefSeq NM_017643. `gene_exact:MBTD1 AND organism_id:9606 AND reviewed:true` returns exactly **one** reviewed row whose primary gene name reads back as MBTD1, with no gene synonyms in UniProt at all; HGNC lists **no previous official symbol** and two aliases, `SA49P01` and `FLJ20055`. Feature table: **FCS-type zinc finger 45-80** and **four MBT repeats at 141-245, 253-350, 351-456 and 464-560**, with disordered stretches at 1-31, 560-590 and 606-628 [E1]. **What it reads and what it is part of.** MBTD1 selectively binds **mono- and di-methylated histone H4 Lys-20 (H4K20me1/me2)** through the fourth MBT repeat, in a semi-aromatic cage using the cavity-insertion recognition mode common to the MBT family [E2][E3]. It is a **stable subunit of the NuA4/TIP60 acetyltransferase complex**, enlisted by the non-catalytic subunit **EPC1**, whose hydrophobic C-terminal fragment grips the MBT repeats at a site **distinct from the H4K20me-binding site** [E3][E4]. Inside the complex, MBTD1's H4K20me binding directs TIP60 to specific gene promoters and promotes **double-strand-break repair by homologous recombination**, partly by competing with 53BP1 for H4K20me2 and by TIP60 acetylating H2AK15 to block the ubiquitylation 53BP1 needs [E3]. **Its normal haematopoietic job is the one that matters for any intervention here.** MBTD1 is highly expressed in fetal HSCs and sustains them; germline-null mice die soon after birth, apparently from skeletal abnormalities. **In conditional knockout adults, HSC and progenitor numbers INCREASE, the HSCs cycle hyperactively, and the response to exogenous stress fails.** MBTD1 binds the *FoxO3a* promoter directly and interacts with TIP60-complex components, and restoring FOXO3a activity in Mbtd1-deficient HSCs rescues both the cycling and the pool-size abnormalities [E5]. So MBTD1 is a **quiescence brake**, and the loss-of-function phenotype is expansion-plus-exhaustion, not aplasia. **It is a fusion partner in two directions.** In AML it is the **3' partner** of `fusion-zmynd11-mbtd1` and supplies the NuA4/TIP60 docking surface [E6]. In **low-grade endometrial stromal sarcoma** it is the **5' partner** of MBTD1::CXorf67, from a reciprocal t(X;17)(p11.2;q21.33) found in three of twenty-eight tumours screened [E7]. That polarity reversal is the same phenomenon this graph already records for KMT2A in sarcoma (`concept-kmt2a-fusion-polarity-is-inverted-in-sarcoma`), in a second gene and a second disease pair.
This is a prediction, not a measurement. No one has seen this protein in this shape. The colours say how much the software trusts itself, and that is worth reading before anything else — the orange stretches are places the model is effectively saying this part has no fixed shape.
I have never read one of these pictures before
A protein is one long chain of amino acids that folds up. The ribbon traces that chain from one end to the other; it is not a wire, it is a simplification of where the backbone runs. Drag to rotate, scroll to zoom.
Domains are the stretches of chain that do a particular job, and they are what the coloured blocks above the picture mark out. Click one and the view flies to it. In this project the important question about any domain is usually whether the fusion still has it — the labels say lost where it does not.
Anything in yellow sticks is a bound small molecule, which is usually the drug.
MBTD1 was filed as a Polycomb repressor and turned out to be a subunit of a coactivator
The 2009 structural paper opens by describing MBTD1 as "one such PcG protein" - a Polycomb-group member, by domain homology to L3MBTL1 and L3MBTL2, and by the family's association with transcriptional repression of developmental control genes 2. Seven years later the same protein was identified as a stable subunit of the NuA4/TIP60 acetyltransferase complex, i.e. of a potent transcriptional coactivator 3.
This matters for two practical reasons. First, any database annotation, GO term or pathway membership for MBTD1 predating 2016 may class it as a repressor, and a coverage query or enrichment analysis keyed on Polycomb membership will pick it up on the wrong side. Second, the assay that assigned it was peptide binding and the assay that reassigned it was complex purification - the same order of events this graph records for several KMT2A partners, where domain homology predicted one thing and the purified complex said another.
The historical framing also explains the deposit list. PDB 4C5I is titled "Crystal structure of MBTD1 YY1 complex" and contains MBTD1 residues (440 modelled, Q05BQ5) with a 30-residue human YY1 peptide (P25490), yet its primary citation is a Drosophila Sfmbt-and-Polycomb-response-element paper 8. It is a side panel in a Polycomb structural study, not an MBTD1 study, and this graph makes no claim about MBTD1-YY1 from it beyond recording that the deposit exists.
The structures, and the one that is not a co-crystal
| PDB | resolution | what it actually is |
|---|---|---|
| 3FEO | 2.5 A | MBTD1 residues 130-566, the four MBT repeats alone, in an asymmetric rhomboid architecture 2 |
| 6NFX | 1.95 A | a SINGLE-CHAIN ENGINEERED CHIMERA. One polymer entity of 470 residues, annotated to both Q05BQ5 (MBTD1) and Q9H2F5 (EPC1) 49 |
| 4C5I | 2.59 A | MBTD1 (440 residues) plus a 30-residue YY1 peptide; primary citation is a Drosophila Sfmbt paper 8 |
6NFX is the load-bearing structure for the fusion mechanism and it is not a measurement of association. RCSB reports polymer_entity_count_protein: 1 and the entity description reads "MBT domain-containing protein 1,Enhancer of polycomb homolog 1" with two UniProt cross-references on the same chain. The paper describes a crystal structure of "the MBTD1-EPC1 complex"; the deposit models it as one fused polypeptide. That is a legitimate and common way to solve a weak or entropically costly interface, and it means the deposit models the bound state rather than demonstrating spontaneous association. The association itself is supported independently by the paper's cellular assays and by the co-purification in 3, so nothing here is in doubt - but an entry or figure describing 6NFX as "MBTD1 bound to EPC1" would be describing a construct.
This graph has made exactly this mistake before, on 2LM0 in interface-aff1-af4int-af9-enl-ahd, which is why the check was run.
Cell type, cell state, and the localisation record - including what UniProt's evidence codes actually say
Cell type. Fetal haematopoietic stem cells (high expression, MBTD1 sustains number and function); adult bone-marrow HSCs and progenitors (conditional knockout target); mouse oocytes during meiotic maturation 10; whole-embryo (germline-null lethality soon after birth with skeletal abnormalities) 5. In cancer: AML blasts with minimal differentiation via the fusion 6, and low-grade endometrial stromal tumours via the reciprocal translocation 7. No human leukaemia measurement of wild-type MBTD1 exists - every haematopoietic result is murine.
Cell state. Quiescence versus cycling, explicitly. Mbtd1-deficient adult HSCs show a hyperactive cell cycle and a defective response to exogenous stress, with more HSCs and progenitors, not fewer, and the mechanism runs through FOXO3a - the canonical HSC quiescence factor 5. In oocytes the state axis is meiotic maturation 10. In the fusion, the state is self-renewal with a differentiation block, but that is a property of the chimera and not of MBTD1.
Subcellular localisation, with the evidence code read rather than filtered. UniProt Q05BQ5 gives two location comments: Nucleus, which is ECO:0000305 - curator inference, NOT an experiment - and Chromosome, which is ECO:0000269:PubMed:27153538 - experimental 13. The stronger-sounding of the two statements is the unsupported one. This is the pattern rule 91 was written about, and it has a real consequence here: the only experimentally coded spatial claim for MBTD1 is chromatin association, which is a chromatin-fractionation-and-ChIP statement, not an imaging one.
Subnuclear localisation: NOT MEASURED. No speckle, PML-body, nucleolar, nuclear-lamina or nuclear-matrix assignment for MBTD1 was found in UniProt or in any of the papers read this session, and none is asserted. The available spatial resolution is genomic: MBTD1-dependent TIP60 occupancy at specific gene promoters 3 and direct binding at the FoxO3a promoter 5.
Epigenetic readout. Reads H4K20me1 and H4K20me2 through MBT4; its presence in NuA4/TIP60 couples that reading to H4 and H2A acetylation and to H2AK15 acetylation; it competes with 53BP1 for H4K20me2; and in mouse oocytes it associates with Pr-Set7/SETD8 (the H4K20 monomethylase) and stabilises H4K20me1 - a reader that protects its own mark 10.
DepMap, with both controls stated and one control failure recorded
Open Targets depMapEssentiality, release year 26 month 06, fetched 2026-09-23. Gene-specific screen counts, not a corpus denominator:
| gene | screens | mean gene effect | median | fraction < -0.5 | fraction < -1.0 |
|---|---|---|---|---|---|
| MBTD1 | 1,258 | -0.353 | -0.342 | 17.9% | 0.24% |
| ZMYND11 | 1,258 | +0.011 | +0.018 | 0.24% | 0% |
| KAT5 | 1,258 | -0.607 | -0.609 | 70.2% | 4.7% |
| EPC1 | 1,258 | -0.336 | -0.314 | 22.7% | 1.4% |
| EP400 | 1,258 | -0.932 | -0.934 | 94.3% | 36.3% |
| MEN1 (in-graph reference) | 1,258 | -0.362 | -0.316 | 28.9% | 6.3% |
| EEF2 (pan-essential control) | 1,258 | -2.709 | - | - | - |
| OR4F5 (low-dependency control) | 5 | -0.043 | - | - | - |
| POLR2A (intended pan-essential control) | 0 | - | - | - | - |
Both controls that returned data behaved correctly, so the endpoint is working. POLR2A returned zero screens through the identical query shape, which is a per-gene gap in this endpoint and not an instrument failure - but it is recorded here because a zero from this endpoint on a single gene is therefore uninterpretable on its own, and a beat that had used POLR2A as its only positive control would have concluded the instrument was dead.
What the numbers say. MBTD1's dependency is real but modest and essentially non-selective (-0.353 mean; the most negative leukaemia line is HNT-34, AML, at -0.764 of 57 leukaemia screens), close to MEN1's own profile. No KMT2A-rearranged-versus-other split was run, so nothing here is a selectivity claim, and this project's own rules on DepMap - lineage-split before stating, a z is not an effect size, floor compression - all apply and none has been applied. ZMYND11 is not a dependency in any line, which is consistent with its wild-type role as a putative tumour suppressor. The two structural nodes of the complex the fusion hijacks, KAT5 and EP400, are near-pan-essential, which is the arithmetic behind this entry's bite-back.
What the fusion does with MBTD1, and the module it does not need
ZMYND11::MBTD1 retains the FCS zinc finger and all four MBT repeats 6. Mutagenesis in murine HSPCs separates them sharply 11:
- delta-MBTx4 (all four repeats deleted) - no Tip60 binding, no transformation
- A661D/L684D/L688D (ALL>DDD, the MBT1-2 residues that grip EPC1, fusion numbering) - no transformation
- delta-ZnF - still transforms, with a reduced phenotype
- W954A and F951A/W954A/Y958A (FWY>AAA), the H4K20me1/2-binding-defective mutants - bind Tip60 normally and transform normally
So the fusion needs MBTD1 as a scaffold and not as a reader. Subtracting the construct offset (ZMYND11 aa 1-409 plus a 16-residue linker, so MBTD1 residue n = fusion residue n + 425), fusion W954 is MBTD1 W529, F951 is F526 and Y958 is Y533 - all three inside MBT4 (464-560) as expected 111. The offset arithmetic is this entry's derivation; the fusion-numbered residues are the paper's.
The consequence is a clean statement about what a reader domain is worth: MBTD1's own biology - HSC quiescence, HR repair pathway choice, 53BP1 competition - runs through H4K20me reading, and the oncogenic chimera keeps the cage and ignores it. See mark-h4k20me1.

| accession resolution | gene_exact:MBTD1 AND organism_id:9606 AND reviewed:true returned exactly ONE reviewed record (Q05BQ5, 628 aa) whose gene_primary reads back as MBTD1, with an EMPTY UniProt gene-synonym field. data/symbol-collisions.json (208 rows) was word-boundary grepped for MBTD1, SA49P01 and FLJ20055 before the first query: no row for any of them. HGNC lists no previous official symbol. Nearest four-MBT-repeat relatives differ in length by 77-238 residues (L3MBTL1 772, L3MBTL2 705, SFMBT1 866), so the length check discriminates here. |
| alphafold ID | AF-Q05BQ5-F1 |
| role | Chromatin reader subunit of the NuA4/TIP60 histone acetyltransferase complex. Binds H4K20me1 and H4K20me2 through the fourth MBT repeat; is enlisted into the complex by EPC1 at a surface distinct from the methyl-lysine cage; directs TIP60 to specific gene promoters; promotes homologous-recombination repair partly by competing with 53BP1 for H4K20me2 and partly through TIP60-mediated H2AK15 acetylation. In mouse haematopoiesis it is a quiescence brake on adult HSCs acting through direct binding at the FoxO3a promoter. In cancer it is a fusion partner in both polarities: 3' in ZMYND11::MBTD1 AML, 5' in MBTD1::CXorf67 low-grade endometrial stromal sarcoma. |
| epigenetic readout | Reads H4K20me1/me2 and converts that reading into H4 and H2A acetylation by the NuA4/TIP60 complex it belongs to, plus H2AK15 acetylation that blocks the RNF168-dependent ubiquitylation 53BP1 requires. Also reported to STABILISE H4K20me1 in association with Pr-Set7/SETD8 in mouse oocytes, which would make it a reader that protects its own mark. In ZMYND11::MBTD1 the cage is retained and DISPENSABLE for transformation; what the fusion uses is the EPC1-binding scaffold surface. |
| subcellular localization | UniProt Q05BQ5 gives Nucleus with evidence code ECO:0000305 - CURATOR INFERENCE, NOT AN EXPERIMENT - and Chromosome with ECO:0000269:PubMed:27153538, which is experimental. The only experimentally coded spatial statement is chromatin association. No imaging study of MBTD1 was found this session. |
| subnuclear localization | NOT MEASURED. No speckle, PML-body, nucleolar, lamina or nuclear-matrix assignment exists in UniProt or in any paper read this session, and none is asserted. Available spatial resolution is genomic: MBTD1-dependent TIP60 occupancy at specific promoters, and direct binding at the mouse FoxO3a promoter. |
| localization shown for fusion | NO - nobody has looked in any fusion. No localisation experiment on MBTD1 or on ZMYND11::MBTD1 in any fusion-driven system was found. |
| cell type | Mouse fetal HSCs (high expression, MBTD1 sustains number and function); mouse adult bone-marrow HSCs and progenitors (conditional knockout); mouse oocytes in meiotic maturation; whole embryo (germline-null lethal soon after birth with skeletal abnormalities). Human disease contexts: minimally differentiated AML blasts via ZMYND11::MBTD1, low-grade endometrial stromal tumours via MBTD1::CXorf67. NO human haematopoietic measurement of wild-type MBTD1 was found. |
| cell state | Quiescence versus cycling. Mbtd1-deficient adult mouse HSCs show a hyperactive cell cycle, increased HSC and progenitor numbers, and a defective response to exogenous stress; FOXO3a restoration rescues both cycling and pool size. Note the direction reverses between fetal life (MBTD1 sustains HSC number) and adult life (MBTD1 loss increases it). |
| shown for fusion | ZMYND11::MBTD1 only, and there as the 3' partner supplying the complex-docking surface - NOT as a reader, since the H4K20me-cage mutants transform normally. Nothing in any KMT2A fusion: no co-immunoprecipitation, no occupancy, no dependency arm. |
| relationship to existing entries | gene-kat5 and gene-ep400 cover the catalytic and ATPase subunits of the same complex and already name MBTD1 in passing; this entry supplies the reader subunit and the EPC1 recruitment arm neither of them holds. gene-zmynd11 (Q119) covers the 5' partner and its open_questions field asks for exactly this entry and for fusion-zmynd11-mbtd1. mark-h4k20me2 covers the dimethyl state and its readers and explicitly records that no 53BP1 or H2AK15ub node exists; this entry adds MBTD1 as a second H4K20me2 reader it does not list, and mark-h4k20me1 (written in the same tranche) covers the monomethyl state it does not hold. |
| literature bound 2026 09 23 | MBTD1[tiab] = 35; MBTD1[tiab] AND fusion[tiab] = 14; MBTD1[tiab] AND TIP60[tiab] = 7; MBTD1[tiab] AND NuA4[tiab] = 5; MBTD1[tiab] AND sarcoma[tiab] = 6; MBTD1[tiab] AND CXorf67[tiab] = 4; MBTD1[tiab] AND hematopoietic[tiab] = 3; MBTD1[tiab] AND H4K20[tiab] = 2; MBTD1[tiab] AND EZHIP[tiab] = 0. All through scripts/pubmed_safe.py; titles and abstracts only. The entire primary literature on this gene is 35 records. |
| kmt2a relationship bound | NOT QUERIED AS A PAIR IN THIS ENTRY. MBTD1[tiab] AND KMT2A[tiab] and MBTD1[tiab] AND MLL[tiab] were NOT run, so no absence is asserted on that pairing. The related-but-different statement that is supported: MBTD1 appears in 35 PubMed records in total, of which none read this session concerns KMT2A, and MBTD1 is named in only two of 2,331 node files in this corpus. |
| absence class | never-measured |
| absence class note | Two arms. (1) The HUMAN WILD-TYPE arm is never-measured: no human haematopoietic experiment on wild-type MBTD1 exists in any form - every HSC result is a mouse conditional knockout - and closing it needs primary human cells or patient material, i.e. wet work. (2) A cheaper arm exists and is deliberately NOT given an accession here: whether MBTD1 or NuA4/TIP60 subunits are differentially expressed or dependent in KMT2A-rearranged versus other leukaemia is computable from DepMap and from public expression compendia, which would make it measured-not-analysed - but the lineage split was not run this session and naming a specific series or portal from memory is the failure this field exists to prevent. The single class above names the arm that dominates the gene's biology. |
| archive check 2026 09 23 | NONE RUN for MBTD1. No GEO, PRIDE, ProteomeXchange or dbGaP query was executed for this gene this session. The absence statements above are therefore bounded by PubMed titles and abstracts plus UniProt, RCSB, HGNC, Ensembl and Open Targets, and by nothing else. Recording the omission rather than implying a sweep. |
| salvaged cycle 82 | Q121 was killed by the account session limit after writing this entry's evidence but before its counter_evidence. The coordinator added 11 reasoned methodological counter-arguments during salvage, each derived from the claim and its citation type and each labelled as such. NO counter-citation was fetched and none is asserted - inventing one would have been the fabrication this project has a rule against. Every note also states what a stronger, literature-sourced counter would be, so the gap is recorded rather than closed. |
| salvage notes withdrawn cycle 86 | Three of the four flags the coordinator attached to this entry during cycle 82's salvage do NOT stand, and are replaced in place rather than deleted so the record shows what was claimed. E12's premise was factually false (the Open Targets API DOES return meta.dataVersion 26.06 - verified live by the coordinator before withdrawing); E8's concern was checked by sequence alignment and the entry was right; E9's note restated the entry's own content. Found by Q123. The salvage notes were written as 'reasoned methodological counter-arguments' with no counter-citation fetched - which is exactly the condition under which three of four turned out wrong. |

}- MBTD1's normal function is a brake. Conditional knockout does not deplete HSCs - it expands them, makes them cycle hyperactively and breaks their stress response. An agent that disables MBTD1, or the EPC1 surface that recruits it, would be releasing that brake in every normal stem cell it reaches. In an infant, whose marrow has to work for eighty years, releasing HSC quiescence is a worse risk than a cytopenia.
- MBTD1 promotes homologous-recombination repair and restrains 53BP1. Disabling it would be expected to shift double-strand-break repair pathway choice towards end joining - which is the pathway this project's break-machinery layer argues produces the KMT2A translocation in the first place. A drug that biases repair towards NHEJ in a haematopoietic progenitor is a drug with a second-malignancy argument attached.
- The complex MBTD1 sits in is near-essential. KAT5 mean gene effect -0.607 with 70.2% of 1,258 lines below -0.5, and EP400 -0.932 with 94.3% below -0.5. 'Inhibit the hijacked acetyltransferase' is an essential-function toxicity proposal. MBTD1 itself is shallower (-0.353, 17.9%), which is the reason to target the reader rather than the enzyme - and also the reason a reader inhibitor may do very little.
- The fusion does not use MBTD1's reader function, so a methyl-lysine-cage ligand against MBT4 would be a chemically tractable drug against the wrong module. This is the mirror of the LEDGF PWWP problem this graph already records: the arm with chemistry is not always the arm with genetics.
- Every haematopoietic statement in this entry is mouse. There is no human MBTD1 loss-of-function phenotype, no human expression atlas reading quoted here, and no patient with MBTD1 haploinsufficiency described. A toxicity argument built on a mouse conditional knockout is a hypothesis about humans.
- Is MBTD1 or any NuA4/TIP60 subunit differentially required in KMT2A-rearranged lines? The DepMap numbers in this entry are unsplit by lineage or fusion status, and the split is compute on data this project already queries. It is the cheapest MBTD1 question in the corpus.
- Does the H4K20me cage matter for LEUKAEMIA MAINTENANCE even though it is dispensable for transformation? The cage mutants were scored in a serial-replating immortalisation assay. A dispensable-for-initiation module can be required for maintenance, and the distinction decides whether MBT4 is a target surface or a bystander.
- Is MBTD1 a reader that protects its own mark? The mouse-oocyte report has it associating with Pr-Set7/SETD8 and stabilising H4K20me1. If that holds in a haematopoietic cell it inverts the usual reader logic and would mean depleting MBTD1 lowers H4K20me1 as well as un-reading it - which the BAHCC1 replication work would care about.
- Why does adult Mbtd1 loss EXPAND the HSC pool when fetal MBTD1 sustains it? The same paper reports both directions. A stage-dependent reversal in a chromatin reader is the kind of thing this project's fetal-versus-adult origin arguments turn on, and the mechanism is unexplained.
- Has anyone put MBTD1 and KMT2A in the same query? MBTD1[tiab] AND KMT2A[tiab] and MBTD1[tiab] AND MLL[tiab] were deliberately NOT run in this beat, so the pairing is unbounded rather than empty. One query and its title check would settle it, and by this project's own registry the KMT2A operand must be run alongside MLL before any zero is believed.

All three records fetched live on 2026-09-23 and the primary gene name read back. Only one reviewed row is returned so the several-rows trap does not fire, and there is no paralogue close in length that an accession swap could hide behind - the nearest four-MBT-repeat relatives L3MBTL1 (772 aa), L3MBTL2 (705 aa) and SFMBT1 (866 aa) are all far longer, and none carries MBTD1 as an alias. The Ensembl coordinates are GRCh38; no GRCh37 coordinates were fetched, so none are quoted.

The crystal structure of MBTD1 residues 130-566, covering the four MBT repeats, at 2.5 A resolution shows an asymmetric rhomboid architecture resembling L3MBTL2, and fluorescence polarization confirms that MBTD1 preferentially binds mono- and di-methyl-lysine histone peptides through one of its four repeats using a semi-aromatic cage.

Read at abstract level plus the RCSB entry for 3FEO. Binding is to histone PEPTIDES by fluorescence polarization, not to nucleosomes; the MBT family is known to show little peptide-sequence selectivity, so this establishes methyl-state preference more firmly than it establishes H4K20 as the physiological ligand - that assignment comes from E3. The paper frames MBTD1 as a Polycomb-group protein, which the later complex purification contradicts.
Evidence that argues against this (1)
- The paper's own framing of MBTD1 as a Polycomb-group transcriptional repressor was overturned by its identification as a stable subunit of the NuA4/TIP60 acetyltransferase complex, a coactivator. MBTD1 is a stable subunit of the NuA4/TIP60 acetyltransferase complex and, through a histone reader domain for H4K20me1/2, allows TIP60 to associate with specific gene promoters and to promote repair of DNA double-strand breaks by homologous recombination; the complex regulates 53BP1 association partly by competing for H4K20me2 and partly by TIP60 acetylating H2AK15, the residue whose RNF168-dependent ubiquitylation 53BP1 requires.

Read at abstract level; no PMCID was pursued. Human cell lines, not haematopoietic primary cells, and the DNA-repair readouts are reporter and focus assays rather than patient material. UniProt cites this paper as the experimental support for both the H4K20me1/2 binding and the Chromosome localisation of Q05BQ5, and it is the paper the function comment is actually about - so this is a case where the database pointer and the source agree, unlike several logged in this project.

In mice, MBTD1 is highly expressed in fetal HSCs and sustains their number and function; germline-null animals die soon after birth, apparently from skeletal abnormalities; and in conditional knockout adults HSC and progenitor numbers increase, HSCs show a hyperactive cell cycle and a defective response to exogenous stress, with MBTD1 binding the FoxO3a promoter directly, interacting with TIP60-complex components, and restoration of FOXO3a activity rescuing both the cycling and the pool-size abnormalities.
