CALM-AF10 reaches DOT1L through exactly the same OM-LZ interface as KMT2A::MLLT10 - the recruitment transfers, the addressing does not
also called CALM-AF10 DOT1L recruitment · OM-LZ interface transfer
Both the KMT2A::MLLT10 and the CALM::AF10 leukaemia fusions keep the same small piece of the AF10 protein, and that piece grabs the DOT1L enzyme in the same way. What differs is how each fusion decides WHERE on the genome to take DOT1L.
The same thing, for a scientist
**The interface is identical.** Both fusions retain the AF10 **octapeptide-motif/leucine-zipper (OM-LZ)**, and it is the same physical contact in both: the AF10 LZ binds the DOT1L coiled-coil domains through a conserved binding mode, and the LZ C-terminus plus the OM drive **tetramerisation of the binary complex into a DOT1L-AF10 octamer** whose oligomerisation is essential for leukaemogenic function [E1]. That was established for MLL1-AF10 and the retained module is the same in CALM-AF10, where the OM-LZ is likewise necessary and sufficient with the CALM C-terminus [E2][E3]. **So the DOT1L half of the analogy transfers completely.** **The addressing does not.** KMT2A::MLLT10 additionally carries the KMT2A N-terminus - AT-hooks, the CXXC CpG-binding module and the **menin-binding motifs** - so its DOT1L is delivered to a menin- and CXXC-defined chromatin address. CALM-AF10 has none of that: its targeting comes from a CRM1-dependent export cycle [E4], from a **MOZ/ENL complex that recruits and retains the AF10 fusion/DOT1L complex on chromatin via MOZ/MORF acetyltransferase activity** [E5], and, in the murine AML model, from CTCF-dependent architecture [E6]. **Naming this split is the point of the entry**: an entry that says 'AF10 fusions recruit DOT1L' is true of both and says nothing about menin; an entry that says 'the KMT2A::MLLT10 mechanism transfers to CALM-AF10' is true of the enzyme recruitment and false of the address.
What is shared and what is not, item by item
| element | KMT2A::MLLT10 | PICALM::MLLT10 |
|---|---|---|
| AF10 OM-LZ retained | yes | yes |
| DOT1L coiled-coil contact | yes, same mode | yes, same mode |
| DOT1L-AF10 octamerisation required | yes 1 | assumed by shared module; not separately demonstrated in CALM-AF10 |
| AF10 PZP reader | lost | lost 7 |
| menin-binding motif | yes | no |
| CXXC / post-CXXC CpG address | yes | no |
| MOZ/ENL-dependent chromatin retention | shown for AF10 fusions | shown, CALM-AF10 specifically 5 |
| CRM1/NES-dependent cycling | no | yes, required 4 |
| dominant lineage | myeloid | T-lymphoid |
The one honest gap in this table is the octamerisation row: it was demonstrated for MLL1-AF10 and is inferred, not shown, for CALM-AF10.
Why this decides whether the analogy is usable
This project's central claim is that the menin contact plus the CXXC address is what makes a KMT2A fusion leukaemogenic. CALM-AF10 keeps the DOT1L recruitment and discards both of those, and still produces the HOXA/MEIS1 programme. So either (a) the menin/CXXC address is not necessary for HOXA activation and merely one of several ways to reach it, or (b) CALM-AF10 reaches the same address by a different route that nobody has mapped. Those are different claims with different consequences, and the experiments that separate them are chromatin-occupancy experiments, not viability experiments. ChIP-seq of CALM-AF10 and of menin in the same cells, asked whether they occupy the same HOXA elements, would do it.
Chemical matter at this interface
Because the interface is shared, an OM-LZ/coiled-coil inhibitor would hit KMT2A::MLLT10 and PICALM::MLLT10 with the same molecule - a rare case where one agent covers a KMT2A fusion and a non-KMT2A one. Structure-optimised inhibitory peptides against the DOT1L-AF10 coiled coil have been reported 1, and crystal structures of the complex exist (6CKN, 6CKO, 6JN2) 8. No drug-like small molecule against this interface was found in this beat's reading.

- An OM-LZ inhibitor would also block the ENDOGENOUS AF10-DOT1L complex, which the CRISPR-ChIP partition assigns to normal cells; DOT1L is required for normal adult haematopoiesis, so this interface is not a tumour-specific one.
- MOZ/MORF KAT inhibition is upstream of a large amount of normal transcription and its therapeutic window is not established in this disease.
- The shared-interface argument is a structural inference for CALM-AF10, not a measurement. If the CALM-AF10 complex turns out to have different stoichiometry, a peptide optimised on MLL1-AF10 may not transfer.
- Does CALM-AF10 form the same DOT1L-AF10 octamer? The requirement was shown for MLL1-AF10 and is assumed here from the shared module. A size-exclusion or crosslinking experiment on the CALM-AF10 complex would close it.
- Do CALM-AF10 and menin occupy the same HOXA elements? This is the experiment that distinguishes 'a different address' from 'the same address reached differently', and it has not been done.
- Would a MOZ/MORF KAT inhibitor work in KMT2A::MLLT10 as well as in CALM-AF10? The recruiting complex is reported for AF10 fusions generally, which would make it a partner-defined rather than a fusion-defined vulnerability.

The AF10 leucine zipper binds the DOT1L coiled-coil domains through a conserved binding mode, and the C-terminal end of the LZ together with the OM mediates tetramerisation of the binary complex into a DOT1L-AF10 octamer whose oligomerisation ability is essential for MLL1-AF10's leukaemogenic function.
A higher-order configuration of the heterodimeric DOT1L-AF10 coiled-coil domains potentiates their leukemogenenic activity. · Song X, et al. · Proc Natl Acad Sci U S A 2019What this evidence cannot show
Structural and functional work framed around **MLL1-AF10**, not CALM-AF10. The retained AF10 module is the same, so the interface transfers by construction; the octamerisation REQUIREMENT has not been separately tested in CALM-AF10.
Evidence that argues against this (1)
- Per the brief's condensate and oligomerisation caveats, a higher-order assembly requirement demonstrated with engineered constructs is not the same as showing the assembly forms at endogenous stoichiometry in a patient cell. Song X et al. 2019

In mice, the C-terminal 248 residues of CALM fused to the AF10 OM-LZ domain are sufficient to induce AML with Hoxa cluster upregulation.

Murine retroviral minimal construct; transforms more efficiently than the full-length fusion, so it is a tool rather than a model of the patient protein.
Evidence that argues against this (1)
- PUBLISHED CONFLICTING RESULT on the attribution, from a mouse model of the same region. This item's title attributes the minimal construct's activity to the CLATHRIN-BINDING domain. The model built to test that attribution found against it: excluding the C-terminal portion of CALM required for optimal clathrin binding produced myeloproliferative disease rather than AML - so the region matters - but the development of leukaemia COULD NOT BE ATTRIBUTED DIRECTLY to interference with endocytosis, full-length fusion showed no consistent effect on growth-factor endocytosis, and the authors propose HOMO-OLIGOMERISATION, measured by FRET, as what endocytic 5' partners actually supply. Sufficiency of the fragment is not in dispute; what the fragment does is. Stoddart A et al. 2012

In murine bone-marrow serial-replating assays, PICALM::MLLT10 lacking the Dot1l-interacting OM-LZ domain cannot sustain colony formation, while wild-type PICALM::MLLT10 continues through the third plating.
Leukaemic transformation by CALM-AF10 involves upregulation of Hoxa5 by hDOT1L. · Okada Y, et al. · Nat Cell Biol 2006What this evidence cannot show
Murine bone-marrow serial colony-plating. **Verified in the paper's full text on 2026-09-20 via the PMC HTML route (PMC4425349), not from the abstract**: the paper states that the capacity to generate a significant number of third-round colonies is dependent on the ability of CALM-AF10 to interact with hDOT1L, with the OM-LZ deletion mutant as the comparator. The same paper's headline mechanism - that hDOT1L contributes by PREVENTING nuclear export of CALM-AF10 - is in tension with later work showing that nuclear export is REQUIRED for transformation; see mechanism-calm-af10-crm1-nuclear-export-requirement.
Evidence that argues against this (1)
- Okada et al. report that hDOT1L contributes to transformation partly by preventing nuclear export of CALM-AF10, i.e. by making the fusion MORE nuclear. Conway et al. later showed that a nuclear-restricted CALM-AF10 does not transform and that export is necessary. Both cannot be straightforwardly true and the graph should not smooth them. CALM residues 544-553 form a CRM1-dependent NES that is necessary for CALM-AF10 transformation, and a bare NES fused to AF10 is sufficient to immortalise murine bone-marrow cells.

Murine bone marrow and HEK293 imaging.
Evidence that argues against this (1)
- PUBLISHED CONFLICTING RESULT: three claims are being made about one piece of protein and none of the experiments separates them. CALM 544-553 is an NES; the same C-terminal span is the clathrin-binding region; and the same span confers a propensity to homo-oligomerise, shown by fluorescence resonance energy transfer. Excluding the clathrin-binding C-terminus changes the mouse disease from AML to a myeloproliferative disease, while the leukaemia itself could not be attributed to interference with endocytosis. So deleting or restoring this region moves at least three variables at once, and 'a bare NES fused to AF10 immortalises bone marrow' is also 'a bare oligomerisation-competent peptide fused to AF10 immortalises bone marrow'. The MOZ/ENL complex recruits the DOT1L/AF10 fusion complex and maintains its chromatin retention through MOZ/MORF acetyltransferase activity; ENL's YEATS-domain interaction with MOZ is critical for CALM-AF10-mediated leukaemic transformation, and combined MOZ/MORF plus DOT1L inhibition cooperatively induces differentiation of CALM-AF10 leukaemia cells.
MOZ/ENL complex is a recruiting factor of leukemic AF10 fusion proteins. · Komata Y, et al. · Nat Commun 2023What this evidence cannot show
The system is AF10-translocation leukaemia including CALM-AF10; the mouse/human split is not resolved from the abstract alone. This is the best available answer to 'what addresses CALM-AF10 to chromatin' and it is an acetylation-reader answer, not a menin answer.
Evidence that argues against this (1)
- PUBLISHED CONFLICTING RESULT about what the combination arm is worth. The cooperativity here is between two tool compounds in mice, and the clinical record for the DOT1L half of it, in the population where the H3K79 rationale is strongest, is thin: pinometostat in 51 adults with advanced acute leukaemia enriched for MLL rearrangement produced complete remission in 2 patients, both with t(11;19), with no maximum tolerated dose established and the authors describing the activity as modest. A MOZ/MORF-plus-DOT1L combination proposal inherits that, and the entry should not carry 'cooperatively induces differentiation' as though the DOT1L component had a clinical foundation. CTCF is required for both initiation and maintenance of CALM-AF10-induced AML in mice; CTCF knockout reduces colony formation, induces macrophage-like differentiation, and acts through TGM2, whose promoter gains H3K27me3 and loses H3K4me3/H3K27ac on CTCF loss.
CTCF Is Essential for the Development and Maintenance of CALM-AF10-Induced Leukemia. · Kuroki Y, et al. · Cancer Sci 2025What this evidence cannot show
Murine CALM-AF10 AML with confirmation of the TGM2 arm in human U-937. A requirement for CTCF is not specific to this fusion - CTCF is required by many things - and the entry treats it as an architectural hint, not as a fusion-specific mechanism.
Evidence that argues against this (1)
- PUBLISHED CONFLICTING RESULT that offers a less fusion-specific reading of the CTCF phenotype. The chromatin signature reported at TGM2 on CTCF loss - gain of H3K27me3, loss of H3K4me3 and H3K27ac - is a polycomb switch, and polycomb is independently required on BOTH sides of this graph's comparison: Bmi1 is indispensable for the development of MLL-AF9 leukaemia in vivo, with Bmi1-null cells showing enhanced differentiation and retaining fewer leukaemia stem cells. A CTCF knockout that induces macrophage-like differentiation through a promoter gaining H3K27me3 may therefore be reporting a general PRC1-mediated differentiation switch that any of these leukaemias would show, rather than an architectural dependency specific to CALM-AF10. The PZP domain of AF10 folds as a single module that recognises H3 residues 22-27 and is abrogated by H3K27 modification, and PZP recognition of H3 is required for H3K79 dimethylation and for proliferation of DOT1L-addicted leukaemic cells.

Structural and cellular work on endogenous AF10. It establishes what the fusion LOSES; whether that loss matters is addressed in mechanism-calm-af10-pzp-loss-is-a-removed-brake.
Evidence that argues against this (1)
- PUBLISHED CONFLICTING RESULT: the PZP requirement has the opposite sign in the fusion. In endogenous AF10, PZP recognition of unmodified H3K27 is REQUIRED for H3K79 dimethylation and for proliferation of DOT1L-addicted leukaemic cells. In CALM-AF10, RESTORING a functional AF10 PZP domain promotes nuclear localisation, is required for chromatin association - and ABOLISHES transformation and leukaemogenesis. The same module is necessary in one context and suppressive in the other. A single statement that 'PZP recognition of H3 is required for H3K79 dimethylation' cannot be carried into the fusion, and the cellular arm of the 2015 work was in KMT2A-rearranged lines rather than in any AF10 fusion. Coordinates are in UniProt P55197-1 (1,068 aa) numbering, which is NOT the frame used by the CALM-AF10 clinical literature - the 2026 review works in a 1,027-residue AF10 frame and Conway et al. in a third (NCBI NP_001182555). Never carry an AF10 residue number between a CALM-AF10 paper and a structure without converting the frame.
Evidence that argues against this (1)
- METHODOLOGICAL DEFEATER: a crystallised span is not a functional boundary, and adding it as a fourth frame makes the numbering problem worse rather than better. The constructs behind 6CKN, 6CKO and 6JN2 were chosen to diffract; the functional limits of the AF10 OM-LZ were set by deletion in a transformation assay, where a defined AF10 fragment fused to a CALM C-terminus suffices to induce AML in mice. There is no reason for a crystallisable span and a minimal transforming span to coincide, and quoting 704-779 or 699-782 as 'the OM-LZ' imports a structural-biology convenience into a claim about what the fusion needs. Deshpande AJ et al. 2011

HOW THE ADDRESSED COMPLEX DOES ITS WORK ONCE DOCKED: CALM-AF10 reaches DOT1L through the same OM-LZ interface as KMT2A::MLLT10, so the CRM1 contact supplies the address while OM-LZ supplies the enzyme.
