HEXIM2 quantitatively compensates for HEXIM1 loss to hold the 7SK-bound P-TEFb pool constant - which makes single-gene knockdown of either a buffered experiment, and is the reason a HEXIM1-only measurement cannot be read as the brake's strength
also called HEXIM2 compensation · q181 HEXIM2
HEXIM1 and HEXIM2 both lock the transcription accelerator into its parked state, and they stand in for each other. Remove HEXIM1 and HEXIM2 takes over so completely that the amount of parked accelerator does not change - so an experiment that removes only one of them will look like nothing happened.
The same thing, for a scientist
gene-hexim2 is in this beat's eight-entry real-gap floor and is the member whose absence is most consequential for experimental design rather than for annotation. Yik and colleagues showed that HEXIM2, like HEXIM1, inactivates P-TEFb through a 7SK-mediated interaction; that the two form stable homo- and hetero-oligomers, most likely dimers, which may nucleate 7SK snRNP formation; that they have distinct expression patterns across human tissues and cell lines; and critically that in HEXIM1-knockdown cells HEXIM2 compensates functionally AND QUANTITATIVELY to maintain a constant level of 7SK/HEXIM-bound P-TEFb. The corpus's own gene-hexim2 title already states the compensation. What it does not carry is any epigenetics annotation, and the reason this matters is not coverage bookkeeping: a buffered node means every single-gene perturbation of the brake published to date is underpowered by construction, and this corpus's gene-hexim1 entry - which DOES carry an epigenetics field - is the covered half of a pair that cannot be interpreted alone.
What was shown
1, all in one paper:
- HEXIM2 possesses the ability to inactivate P-TEFb and suppress transcription through a 7SK-mediated interaction, like HEXIM1.
- HEXIM1 and HEXIM2 form stable homo- AND hetero-oligomers, most likely dimers, which may nucleate 7SK snRNP formation.
- The two show distinct expression patterns across human tissues and cell lines -- so which paralogue dominates is tissue-dependent, and nobody has measured it in a KMT2A-rearranged blast.
- In HEXIM1-knockdown cells, HEXIM2 compensates functionally and quantitatively to maintain a constant level of 7SK/HEXIM-bound P-TEFb.
The authors' framing is a tightly regulated cellular process maintaining the balance between active and inactive P-TEFb.
The consequence this corpus should care about
A quantitatively buffered node makes single-gene perturbation uninformative about the pathway. Knock down HEXIM1 and the bound P-TEFb pool does not move -- not because the brake does not matter, but because the other paralogue absorbs the loss. Any experiment, CRISPR screen row or DepMap dependency score for HEXIM1 alone is therefore measuring a compensated system.
This has a direct bearing on how the corpus reads its own coverage. gene-hexim1 carries an epigenetics field; gene-hexim2 carries nothing. The covered member of the pair is the one whose individual measurements are least interpretable. That is the sharpest illustration in this beat of the point made in ana-q181-the-index-fusion-core-has-the-content-and-lacks-the-field: coverage is tracking which protein a beat was dispatched on, not which protein carries the information.
The experiment the pairing implies is a double perturbation, and this corpus records no HEXIM1/HEXIM2 double knockdown or knockout in any leukaemia system.

]- Any therapeutic logic that runs through 'strengthen the 7SK brake to reduce elongation at fusion targets' has to contend with a node that is quantitatively buffered by a paralogue. A compound acting on HEXIM1 alone would be expected to show a flat dose-response on bound P-TEFb, and would be abandoned as inactive when the mechanism is actually being compensated - a false negative at the screening stage, not at the clinic.
- The reverse also holds and is the more dangerous read: a resistance mechanism running through HEXIM loss would not show up in a single-gene CRISPR screen, because either paralogue's loss is absorbed. If P-TEFb availability buffers menin-inhibitor or DOT1L-inhibitor response, the relevant screen is a paralogue-pair screen and none has been run in this disease.
- gene-hexim1 carrying an epigenetics field while gene-hexim2 carries none would let a reader filtering the graph on this dimension see the buffered half of an obligate pair and not the buffer. That is a navigational hazard created by the coverage gap itself, independent of any biology.
- What is the HEXIM1:HEXIM2 ratio in SEM, RS4;11 and primary KMT2A::AFF1 blasts? The paralogues have distinct tissue expression and nobody has measured either in this disease, so which one is the brake here is unknown.
- Does the reciprocal compensation hold - does HEXIM1 rise on HEXIM2 knockdown? The 2005 paper tests one direction only, and the experimental-design argument in this entry assumes symmetry it does not have.
- Run the bare-operand control on HEXIM2 per Rule 72. Two natural queries returned 0 and 1 records respectively, which is the signature of a name-list problem rather than a small literature.
- Is there a HEXIM1/HEXIM2 double knockout in any system, and does the bound P-TEFb pool collapse? If it does not, there is a third buffer nobody has named.

HEXIM2, a protein highly homologous to HEXIM1, inactivates P-TEFb to suppress transcription through a 7SK-mediated interaction; HEXIM1 and HEXIM2 form stable homo- and hetero-oligomers, most likely dimers, which may nucleate 7SK snRNP formation; the two exhibit distinct expression patterns in human tissues and established cell lines; and in HEXIM1-knocked-down cells HEXIM2 compensates functionally and quantitatively for the loss of HEXIM1 to maintain a constant level of 7SK/HEXIM-bound P-TEFb.
Compensatory contributions of HEXIM1 and HEXIM2 in maintaining the balance of active and inactive positive transcription elongation factor b complexes for control of transcription · Yik JHN, Chen R, Pezda AC, Zhou Q · Journal of Biological Chemistry 280(16):16368-76 2005What this evidence cannot show
Abstract-level read; the full text was not retrieved. HeLa cells; the compensation experiment is a single-direction knockdown of HEXIM1 with HEXIM2 measured, and the reciprocal experiment (HEXIM2 knockdown, HEXIM1 measured) is not described in the abstract. 2005, so the dimer stoichiometry has probably been revised by later structural work that was not searched for. The oligomer claim is hedged in the source ('most likely dimers', 'may nucleate') and both hedges are preserved. expected_absent_terms records that this is not a chromatin paper, which is part of why the entry carries a bounded absence.
Evidence that argues against this (2)
- No paper disagreeing with HEXIM1/HEXIM2 compensation was found, but the search behind that statement was narrow - PubMed 'HEXIM2 cloning characterization P-TEFb 7SK inhibition' returned 0 and 'HEXIM2 HEXIM1 heterodimer 7SK P-TEFb' returned exactly 1 record, this one. A symbol with a single retrievable record under its most natural queries is a symbol whose literature has not been surveyed, and per this project's Rule 72 the bare-operand control ('HEXIM2' alone) was NOT run. The absence of contradiction here should be read as unchecked.
- The 'quantitatively compensates' claim is the load-bearing one for this entry's experimental-design argument, and it rests on a Western-blot-scale measurement of bound P-TEFb in one cell line in 2005. 'Constant' at that resolution could conceal a substantial change - a 30% shift in bound pool would very likely have been called constant - which would weaken but not remove the conclusion that single-gene perturbation is buffered.
