SELE (E-selectin) - the vascular-niche chemoresistance receptor with a clinical antagonist, zero KMT2A papers, and a paediatric trial that a company restructuring closed
also called E-selectin · ELAM1 · CD62E · endothelial leukocyte adhesion molecule 1
E-selectin is a sticky protein on marrow blood vessels that leukaemia cells grab to survive chemotherapy, and a drug that blocks it doubled survival in mice with leukaemia while sparing normal stem cells. Nobody has ever studied it in KMT2A-rearranged leukaemia, and the only trial of that drug that would have accepted a one-year-old was stopped because the company restructured.
The same thing, for a scientist
SELE (UniProt P16581, 610 aa, C-type lectin plus EGF-like plus six Sushi domains) is the endothelial adhesion molecule that this graph's own cycle-40 rejection ledger named as **'the best single target this ledger identifies for a future beat'**, on the strength of a clean zero against a large literature. That zero is re-measured and holds: `(SELE[tiab] OR E-selectin[tiab] OR uproleselan[tiab])` intersected with the KMT2A/MLL half returns **0**, with valid translation and 'No items found.', against an E-selectin operand of 8,608 [E3]. What makes the zero worth a whole entry is what sits on the other side of it. AML blasts release inflammatory mediators that upregulate endothelial niche E-selectin; oncogenesis-associated glycosylation changes enhance blast binding; binding drives pro-survival AKT/NF-kB signalling; blasts with the highest E-selectin binding potential are **12-fold more likely to survive chemotherapy** and are the main contributors to relapse; and absence of E-selectin in *Sele-/-* hosts or blockade with the small-molecule mimetic uproleselan dampens regeneration, strongly synergises with chemotherapy, doubles mouse survival over chemotherapy alone, and **protects endogenous HSCs** [E1]. That last clause is the therapeutic-index property CXCR4 antagonism does not have.
No three-dimensional model is available for this protein, so the map above is the whole picture. That is itself informative: AlphaFold does not model chains this long, and the parts that have been crystallised are held under the entries for the individual domains.
The clinical route existed, reached infants, and was closed for corporate reasons
Three registry facts, all fetched from the raw ClinicalTrials.gov v2 API on 2026-09-22 so that whyStopped was visible:
- NCT05569512, phase 1/2, uproleselan with high-dose busulfan pre-transplant conditioning for chemotherapy-resistant AML, minimum age 12 MONTHS - the only uproleselan study in the registry that reaches an infant. TERMINATED at n=1.
whyStopped: 'On 7/26/24 the Sponsor-Investigator was notified GlycoMimetics was terminating contracting for NCT05569512 following company restructuring. This notification came ahead of the study meeting criteria to progress from Phase 1 to Phase 2.' 4 - NCT05054543, phase 3 bridging trial in Chinese R/R AML, n=140, TERMINATED.
whyStopped: 'In light of the strategic realignment of the company's development focus, it has been determined to discontinue the further development of Uproleselan Injection.' 5 - NCT03701308, NCI randomised phase 2/3 in older adults (60+), n=267, ACTIVE_NOT_RECRUITING 6.
Neither termination is a safety or futility signal, and both strings are invisible to PubMed. The paediatric arm of this mechanism was closed by a sponsor's contracting decision before the study reached its own phase-2 criterion. For a lab choosing between niche targets, that is a different kind of obstacle from the one facing CXCR4 antagonism - and a more tractable one, because the drug and the safety database exist.
Why the zero is worth recording rather than embarrassing
A zero against 8,608 E-selectin papers and 2,922 KMT2A/MLL papers is an absence of intersection, not an absence of literature on either side, and this graph's standing rule is that two large sets with an empty intersection is a claim needing its own evidence. The rule-67 control is in place: the identical query shape with LEPR and leptin in place of the selectin terms returns 2 records, and with SPP1/osteopontin returns 4, so the AND composes and the zero is not an instrument artefact 3.
The prior beat's disposition therefore still holds and is now sharper. Cycle 40 rejected E-selectin outright for having no KMT2A literature and simultaneously flagged it as the best future target. Both halves remain true two cycles later: the literature is still empty, and the preclinical AML case 1 plus the existing clinical agent make this the cheapest well-formed experiment the niche layer can propose - test uproleselan in a KMT2A-rearranged, ideally KMT2A::AFF1, xenograft with chemotherapy, and measure E-selectin-binding potential on the blasts.
| gene symbol | SELE |

| accession resolution | gene_exact:SELE AND organism_id:9606 AND reviewed:true returned exactly ONE reviewed human record, P16581 / LYAM2_HUMAN, primary gene name read back as SELE, 610 aa, synonym ELAM1. Fetched live 2026-09-22. |
| localization | CELL MEMBRANE, two ECO:0000269 supports on UniProt P16581, on the ENDOTHELIAL cell - not on the blast. This is the second molecule in this tranche (with VCAM1) whose compartment is the vessel wall rather than the leukaemia, and the consequence is the same: the leukaemic contribution is on the ligand side, which for E-selectin is a set of sialofucosylated GLYCANS on PSGL1, CD44 and related carriers rather than a protein a gene entry can name. E-selectin is inducible rather than constitutive, and E1's mechanism begins with blasts inducing it. |
| subnuclear localization | NOT APPLICABLE BY TOPOLOGY, argued from architecture. A type I membrane protein with a 32-residue cytoplasmic tail, 535 of 610 residues extracellular, no chromatin-binding module. This field does NOT claim nuclear E-selectin was sought and excluded; no such search was run and a curated record's silence is not evidence. |
| subnuclear absence class | not-applicable-by-topology |
| cell type | THE MARROW SINUSOIDAL ENDOTHELIAL CELL, which this graph holds as cell-type-sinusoidal-endothelial-cell. E-selectin is an inducible endothelial adhesion molecule and the leukaemia's role in E1's mechanism is to INDUCE it via released inflammatory mediators, then to bind it via altered surface glycosylation. No KMT2A-rearranged cell type has ever been assayed for E-selectin binding. |
| cell state | A CHEMORESISTANT, RELAPSE-SEEDING BLAST STATE defined operationally by binding potential: in vivo, AML blasts with the highest E-selectin binding potential were 12-fold more likely to survive chemotherapy and were the main contributors to relapse [E1]. This is the most directly MRD-relevant cell-state definition in this graph's niche layer, because it is a sortable property rather than an inferred one. |
| epigenetic readout | NOT MEASURED AND NOT MEANINGFULLY MEASURABLE ON THE BLAST, because the gene is not expressed there. SELE transcription is an endothelial NF-kB-driven inflammatory response, and in a KMT2A-rearranged patient the endothelium does not carry the fusion - so there is no fusion-driven chromatin question at this locus in the leukaemic cell. The question that IS open and is epigenetic in the broad sense is on the ligand side: the oncogenesis-associated GLYCOSYLATION changes that enhance binding [E1] are set by glycosyltransferase expression (FUT4, FUT7, ST3GAL4/6), and whether any of those is under KMT2A-fusion control has never been asked. ABSENCE CLASS for the glycosyltransferase question: never-measured. |
| shown for fusion | NO, in the strongest available sense of that word: nobody has looked in any KMT2A fusion. (SELE[tiab] OR E-selectin[tiab] OR uproleselan[tiab]) intersected with the KMT2A/MLL half returns 0 with valid translation, against an 8,608-record E-selectin literature and rule-67 controls of 2 and 4 through the identical shape [E3]. The supporting AML mouse work [E1] does not state its driving lesion, so it cannot be claimed as a KMT2A result either. |
| intact union membership | ABSENT from the 277-partner IntAct REST union of the six core baits (analyses/interactome-union-migrated-to-rest/results.json, generated 2026-09-22), checked on the symbol and on P16581. |
| clinical agent | Uproleselan (GMI-1271), a small-molecule E-selectin antagonist. Registry state 2026-09-22: NCT03701308, NCI randomised phase 2/3 in AML patients aged 60+, n=267, ACTIVE_NOT_RECRUITING; NCT05054543, phase 3 bridging trial in Chinese R/R AML, n=140, TERMINATED for commercial discontinuation; NCT05569512, phase 1/2 with pre-transplant conditioning, MINIMUM AGE 12 MONTHS, TERMINATED at n=1 after the sponsor terminated contracting following company restructuring, ahead of the study meeting its own phase-1-to-phase-2 criterion. Neither termination cites safety or futility. |
| collision note | NEW COLLISION, MEASURED HERE, NOT PREVIOUSLY IN THE REGISTRY (read 2026-09-22, case-sensitive over d['collisions'], 177 rows: no row for SELE, E-selectin, uproleselan or P16581). A bare substring grep for SELE over data/graph/nodes returns 89 FILES, which reads as a well-covered gene. A word-boundary grep returns ONE file. The 89 are SELECTIVE (52 occurrences), SELECTIVITY (36), SELEX (15), SELECTED (12), SELECTIVELY (7), SELECTION (3), SELECTS and SELECT - this corpus writes in emphatic uppercase and 'SELECTIVE' is one of its most common words. This is the same family as the registry's WAS, RPE and CPM rows: a short symbol that is a prefix of an ordinary English word. RULE: grep this symbol with -w, or search it as E-selectin. |
| prior beat disposition | ana-marrow-niche-rejected-candidates (cycle 40) rejected E-selectin outright with intersection 0 against a niche half of 8,919, and simultaneously called it 'the best single target this ledger identifies for a future beat'. Re-measured here 2026-09-22: the intersection is still 0 and the E-selectin operand is 8,608 by this beat's own query (E-selectin[tiab] alone; the earlier 8,919 used a wider selectin term). BOTH HALVES OF THE EARLIER DISPOSITION STAND, and this entry converts the flag into a written target rather than overturning it. |

}- THE THERAPEUTIC INDEX CLAIM IS THE WHOLE APPEAL AND IT IS ONE CLAUSE OF ONE ABSTRACT. E1 reports that blockade protects endogenous HSCs while sensitising blasts - the opposite of the CXCR4 problem. That is the reason to pursue this target and it rests on a single unreplicated statement whose figures were not read.
- E-SELECTIN IS AN INFLAMMATORY RESPONSE MOLECULE. Blocking endothelial leukocyte adhesion in a neutropenic infant on intensive chemotherapy impairs neutrophil recruitment to infection sites, and infection is a leading cause of death in infant ALL therapy. No paediatric safety database for this agent exists beyond a single terminated patient.
- THE LIGAND, NOT THE RECEPTOR, MAY BE ABSENT IN THIS LINEAGE. If KMT2A::AFF1 B-precursor blasts do not carry the sialofucosylated glycans, the target is simply not engaged and the whole mechanism is a myeloid one. This is a failure mode that a mouse AML model cannot reveal and that one flow experiment would.
- THE SUPPLY ROUTE IS CLOSED. Both terminated uproleselan trials stopped for commercial reasons, one of them the only protocol that would have admitted a one-year-old. A preclinical programme built on this agent must confirm material availability before it starts.
- Do KMT2A-rearranged B-ALL blasts bind E-selectin at all? The mechanism in E1 is glycan-dependent and the sialofucosylated ligands are best characterised on myeloid cells. One flow-cytometry experiment with an E-selectin-Ig chimera on infant KMT2A::AFF1 samples would settle whether this target is even available in the index disease, and it is the cheapest decisive experiment in this beat.
- Are the fucosyltransferases and sialyltransferases that build E-selectin ligands (FUT4, FUT7, ST3GAL4, ST3GAL6) under KMT2A-fusion control? That would convert a general AML niche mechanism into a fusion-specific one and is answerable from existing KMT2A-r expression data.
- Is uproleselan obtainable for academic preclinical work now that its sponsor has deprioritised it? The scientific case is intact and the commercial one is not, which is a specific and answerable practical question.

In AML mouse models, blasts release inflammatory mediators that upregulate endothelial niche E-selectin; oncogenesis-associated cell-surface glycosylation changes enhance blast binding to E-selectin and promote pro-survival AKT/NF-kB signalling; in vivo the blasts with highest E-selectin binding potential are 12-fold more likely to survive chemotherapy and are the main contributors to relapse; and absence of E-selectin in Sele-/- hosts or blockade with the small-molecule mimetic GMI-1271/uproleselan inhibits this niche-mediated pro-survival signalling, dampens blast regeneration, strongly synergises with chemotherapy, doubles the duration of mouse survival over chemotherapy alone, and protects endogenous HSCs.

AML mouse models generally - the abstract does not name the driving lesion, so whether any arm is KMT2A-rearranged is unknown and must not be assumed (rule 73 in its strictest form: the system is unstated). The 12-fold figure is a between-fraction comparison of binding potential, not a treatment effect. 'Protects endogenous HSC' is the most therapeutically important clause in the abstract and is the one most in need of the figures, which were not opened (rule 80). Read from the abstract.
Evidence that argues against this (1)
- A mechanism that depends on oncogenesis-associated glycosylation changes may not transfer between lineages: the E-selectin ligands on a myeloid blast (sialyl-Lewis-x on PSGL1 and CD44 glycoforms) are not necessarily present on a B-precursor blast, and infant KMT2A::AFF1 disease is B-lineage with a characteristically immature immunophenotype. No E-selectin binding measurement exists on any KMT2A-rearranged B-ALL sample. NCBI PubMed 2026

The two cell-membrane location terms carry ECO:0000269. E-selectin's ligand engagement is carbohydrate-dependent through the C-type lectin domain, so the protein sequence alone does not predict which cells it binds - the glycosylation state of the counterpart cell does, which is the mechanism E1 turns on.
Evidence that argues against this (1)
- A curated feature table records what curators accepted, not what was sought and excluded, so no absence claim follows from it. Rule 67 controls were run through the IDENTICAL query shape with the same disease half: (LEPR[tiab] OR leptin[tiab]) returns 2, (SPP1[tiab] OR osteopontin[tiab]) returns 4, VCAM1[tiab] returns 2 - so the AND composes and this is a real zero. Bounded over titles and abstracts (rule 80): an E-selectin binding assay inside a supplementary figure of a KMT2A paper would be invisible. The name list was SELE, E-selectin and uproleselan; CD62E, ELAM1 and GMI-1271 were NOT searched and are the way this bound could be too tight. Queried 2026-09-22.
Evidence that argues against this (1)
- The hyphenation trap measured elsewhere in this beat applies to this family too: VCAM1[tiab] returns 2 records against the same disease half while VCAM-1[tiab] returns 0. 'E-selectin' was searched in its hyphenated form here, which is the common one, but 'E selectin' unhyphenated and 'CD62E' were not, so the zero is bounded by a three-name list and says so. The 12-month minimum age makes this the only uproleselan record in the registry that would have admitted an infant, and it is an AML transplant-conditioning protocol rather than an infant ALL protocol.
Evidence that argues against this (1)
- A single-patient terminated study is thin evidence that a paediatric route to this mechanism is genuinely available: the study also had to compete for accrual in chemotherapy-resistant paediatric AML, and the CXCR4 precedent in this beat (NCT01655875, terminated at n=1 for lack of enrollment) shows how that usually ends. A commercial discontinuation statement is not an efficacy result and this entry does not read it as one - but it is also not nothing: sponsors sometimes deprioritise after seeing unblinded data, and the registry string cannot distinguish that from a portfolio decision.

- The NCI-sponsored randomised phase 2/3 in older adults, NCT03701308, remains ACTIVE_NOT_RECRUITING with 267 participants, so the mechanism is not clinically dead and a definitive randomised readout may still appear. Evidence that argues against this (1)
- The same monoculture assay returns a flat null for CXCR4 while an in vivo syngeneic MLL-AF9 CRISPR screen made CXCR4 a top cell-surface dependency whose deletion eradicated leukaemia - a direct demonstration that a DepMap null for a niche gene is uninformative about in vivo requirement.
