IGF2BP2 — the paralogue co-over-expressed with IGF2BP3 in MLL/AF4 B-ALL, and the one with an inhibitor in another leukaemia
also called IMP2 · IMP-2 · p62 · VICKZ2 · insulin-like growth factor 2 mRNA-binding protein 2
IGF2BP2 is the sibling of IGF2BP3, and in children with MLL/AF4 leukaemia both siblings are turned up together. That matters because if they do the same job, removing only one of them would hide how important the pair is.
The same thing, for a scientist
IGF2BP2 is one of three IMP-family m6A readers sharing domain architecture and substrate preference with IGF2BP3. In B-ALL, MLL/AF4-positive disease is characterised by over-expression of **both** IGF2BP2 and IGF2BP3, while IGF2BP1 instead marks ETV6/RUNX1 and IGF2BP2 is *under*-expressed in E2A/PBX1 disease [E1]. Functionally it is established elsewhere in leukaemia: IGF2BP2 regulates glutamine metabolism and is a therapeutic target in AML [E2], and its inhibition is a proposed strategy in T-ALL [E3]. Against MLL or KMT2A directly the literature is two records over three aliases [E1]. DepMap gives it a Chronos mean of -0.269 with 4.6% of 1,208 lines depleted and a B-lymphoid index-arm difference of -0.123 against 43 other leukaemia lines - the second-largest negative lymphoid difference among the three paralogues, matching the expression data [E4].
Why this is the paralogue that matters for the IGF2BP3 question
gene-igf2bp3 raises a question it cannot answer: if the family is redundant, single-gene CRISPR under-reports all three, and IGF2BP3's modest Chronos (-0.137, 1.6% of lines) would be explained without the mouse biology being wrong.
The expression data pick a side. In B-ALL entities, the pair that moves up together in MLL/AF4-positive disease is IGF2BP2 and IGF2BP3 1. IGF2BP1 is the ETV6-RUNX1 marker and is low or negative in normal marrow and in this subtype.
This graph's own dependency scan independently picks the same two: lymphoid index-arm differences of -0.123 (IGF2BP2) and -0.135 (IGF2BP3) against +0.095 for IGF2BP1 4. Two datasets generated by entirely different means agree on which two of three paralogues belong to this disease. That is a weak result in each dataset alone and a more interesting one taken together.
It does not establish redundancy. Co-expression is what a redundancy model predicts but it is equally what a shared upstream regulator predicts, and the Marschalek-lab observation that MLL-AF4 induces RNA-binding-protein genes as a counter-regulatory response supplies exactly such a regulator.
What IGF2BP2 does in leukaemia, in other lineages
The functional work is myeloid and T-lymphoid, not KMT2A::AFF1.
In AML, IGF2BP2 regulates glutamine metabolism and is presented as a therapeutic target 2. In T-ALL, inhibition of IGF2BP2 is proposed as a strategy in its own right 3. Both are reader-function stories in the m6A sense, and both are in diseases the index patient does not have.
This graph has repeatedly had to strike myeloid mechanisms imported into infant B-lymphoid disease, so the transfer is recorded as unmade rather than assumed.

| subnuclear localization | NOT APPLICABLE IN PRACTICE, RECORDED AS A BOUNDED ABSENCE, and the reason is a measured one rather than a topological one: the KH domains ACTIVELY PREVENT nuclear accumulation of IGF2BP2, and disrupting all four is what lets the protein into the nucleus [E_q93_igf2bp2_loc3]. So the wild-type protein's nuclear pool is small by design, which is why UniProt's 'Nucleus' value has an empty evidence list while its granule values are experimental. That is not a category exclusion in the sense used for HLA-B, FUT9, B2M or CD33 - nothing structural bars entry, and a mutant enters - so it belongs in the denominator of undetermined subnuclear questions rather than outside it, and the honest record is: no subnuclear compartment assigned, and the protein is excluded from the nucleus by an active, domain-encoded mechanism. Bound: the gene clause against a twelve-term subnuclear clause returns 7 records and ALL SEVEN ARE OFF TOPIC, every one of them matched through 'nucleolar' inside SMALL NUCLEOLAR RNA or an SNHG host-gene name [E_q93_igf2bp2_loc4]. |
| localization shown for fusion | NO - nobody has localised IGF2BP2 in any fusion, in any lineage, although EXPRESSION is documented in the index fusion. The gene clause over IGF2BP2, IMP2, IMP-2 and VICKZ2 returns 1,388 records, against a nine-term KMT2A/MLL fusion clause returns 2 (PMIDs 25537453 and 21414819) and against a localisation clause AND the fusion clause returns ZERO [E_q93_igf2bp2_loc4]. The unasked question is the one this entry was written for: if IGF2BP2 and IGF2BP3 are co-over-expressed in MLL/AF4 B-ALL and are assumed redundant, the paralogue-resolved localisation result says they are NOT interchangeable - all four KH domains govern IGF2BP1 and IGF2BP2 localisation and granule recruitment while IGF2BP3 is only modestly affected - so a redundancy test should read compartment and granule partition, not only knockdown viability. |

}- IGF2BP2 is high in healthy bone marrow and peripheral blood by the same assay that finds it high in MLL/AF4 disease. That is the opposite of the oncofetal profile that makes IGF2BP3 attractive, and it is a direct toxicity argument against pan-family inhibition.
- Near-identical domain architecture across the three paralogues means a selective inhibitor is a hard medicinal-chemistry problem, and a non-selective one inherits IGF2BP2's normal-tissue expression.
- A protein nominated as a therapeutic target in AML, in T-ALL and in MLL/AF4 B-ALL is more likely reporting a general proliferation requirement than a subtype vulnerability.
- Is IGF2BP2 required in KMT2A::AFF1 leukaemia-initiating cells, a question no published study asks?
- Does IGF2BP2 rise when IGF2BP3 is deleted in an MLL-Af4 model, which would be the direct test of compensation?

Evidence that argues against this (2)
- High IGF2BP2 in healthy marrow and blood is the awkward half of this result. A paralogue that is already the abundant family member in normal haematopoietic tissue is a poor candidate for a selective target, and it weakens any argument that IGF2BP2 elevation in MLL/AF4 disease is oncofetal re-expression in the way IGF2BP3's is. Abstract-level. AML, not KMT2A-rearranged B-lymphoid disease. A metabolic mechanism, distinct from the mRNA-stability and splicing operon reported for IGF2BP3 in MLL-Af4.

A statement about the record. Two empty evidence lists on one comment is the third instance this beat has logged after MAP3K14 and JAK1, and here it is instructive rather than alarming: the unsourced values are the vague ones (Nucleus, Cytoplasm) and the sourced ones are the specific compartments, which is the opposite of the DNMT3A pattern where the specific statement was the uncoded one. None of the evidence is haematopoietic.
Evidence that argues against this (1)
- The empty-evidence 'Nucleus' value looks like a defect and is arguably the most defensible line on the record, because a separate paper shows the nuclear pool is ACTIVELY SUPPRESSED: all four KH domains prevent nuclear accumulation of IGF2BP2, and only when all four loops are mutated does the protein accumulate there. So a nucleus value with no experimental citation is describing a compartment the wild-type protein is kept out of - which is real, mechanistically understood, and impossible to cite cleanly as a location. Bounded by the abstract. This is NOT a claim that the compartment data are absent from the paper - 'storage under stress conditions' strongly implies granule imaging in the figures, and UniProt's curator evidently found it. What is established is that the measurement is invisible to any title-and-abstract query, which is why this entry's bounded absence is stated as 'zero in titles and abstracts' per brief rule 80. Human cell lines; nothing haematopoietic.
Evidence that argues against this (1)
- This is the tranche's demonstration, by measurement rather than assertion, that brief rule 80 is the binding constraint on a localisation beat. The bounded query IGF2BP2 AND (stress granule OR P-body) returns two records and UniProt's own compartment citation is not one of them - so the single paper a curator judged sufficient to establish two compartments cannot be found by the query designed to find compartment papers. Every bounded absence written by this beat and its predecessors inherits that limitation, and the correct response is not to widen the name list, which cannot reach a figure, but to open the founding paper for the mechanism, as rule 80 prescribes. The localisation readout comes from LOSS-OF-FUNCTION KH mutants in full-length proteins, so it establishes what the domains do rather than giving a quantified wild-type partition, and no nuclear-to-cytoplasmic ratio is reported. Bounded by the abstract.
Evidence that argues against this (1)
- This result is the strongest available evidence AGAINST this entry's own framing, and the entry should now hold both. The entry exists to record that IGF2BP2 and IGF2BP3 are co-over-expressed in MLL/AF4 B-ALL and to warn that removing only one would hide the pair's importance - a redundancy argument. But on the domain-resolved localisation measurement IGF2BP2 groups with IGF2BP1 and NOT with IGF2BP3: the KH domains govern IGF2BP1 and IGF2BP2 localisation and granule recruitment, and barely affect IGF2BP3. Redundancy at the level of expression pattern and domain architecture does not survive at the level of where the proteins go, so a redundancy test in this disease should read compartment and granule partition rather than knockdown viability alone. Bounded 2026-09-21: the IGF2BP2 gene clause returns 1,388 records; against a twelve-term subnuclear clause it returns 7 and ALL SEVEN are off topic, every one matched through 'nucleolar' inside SMALL NUCLEOLAR RNA or a small-nucleolar-RNA host-gene name; IGF2BP2[tiab] against a P-body and stress-granule clause returns 2, of which one is the domain-resolved localisation paper and UniProt's own compartment citation is NOT present; against a nine-term KMT2A/MLL fusion clause the gene clause returns 2, neither a localisation measurement; and against a localisation clause AND the fusion clause it returns 0.

Titles and abstracts only; supporting figures not searched (brief rule 80), and this entry is the tranche's worked demonstration that the abstract bound is the operative limit rather than a formality. The 1,388-record operand control is inflated by IMP2 and IMP-2 as short aliases and was not title-screened.
Evidence that argues against this (1)
- The 7-of-7 contamination result argues for adding a registry row rather than for distrusting the count, and the direction of the error is worth being precise about. the nucleolar token does not produce a false ZERO - it produces a false POSITIVE literature, seven papers that look like subnuclear hits and are about snoRNAs and their host genes. Under brief rule 94's taxonomy that is the dangerous direction: a zero obtained through a contaminated token stays conservative, but a beat that reported seven subnuclear records for IGF2BP2 without title-checking would have overstated a positive count. The checked number is zero and it is the one recorded. Wächter K et al. 2013
IMP-family paralogue; co-over-expressed in MLL/AF4-positive B-ALL
- co-occurs with →KMT2A::AFF1 (MLL-AF4), t(4;11)(q21;q23)
Over-expressed in MLL/AF4-positive B-ALL alongside IGF2BP3
Second-largest negative B-lymphoid index-arm difference among the three paralogues
The proposed buffering partner
Second single arm and the double arm
PARALOGUE-RESOLVED AND THE PARALOGUES DIFFER: all four KH domains PREVENT nuclear accumulation of IGF2BP1 and IGF2BP2 and FACILITATE their stress-granule recruitment, while the same mutations only modestly affect IGF2BP3 (PMID 23640942). So IGF2BP2 groups with IGF2BP1, not with IGF2BP3 - which cuts against the redundancy premise this entry was written on (Q93 2026-09-21)
Same condensate architecture reached by two protein families: constitutive residence in cytoplasmic mRNP storage granules, stress-INDUCED relocation to stress granules, and a nuclear pool that is either actively excluded (IGF2BP2, by its KH domains) or carrier-dependent (DDX6, piggy-back on 4E-T). Neither measured in a leukaemic cell (Q93 2026-09-21)
Stress-granule recruitment of IGF2BP2 has a MAPPED STRUCTURAL REQUIREMENT - the four KH loops (PMID 23640942) - which is a stronger form of condensate evidence than hexanediol sensitivity or FRAP, the two assays this node records as unable to separate phase separation from clustered-site binding (Q93 2026-09-21)
